4. Mycoplasma Identification Agar Medium
This product is intended for identification of UU&MH mycoplasma in clinic.
The product is used to confirm the existence of urogenital mycoplasma with microscopy at lower magnification, observed with which colonies of Ureaplasma urealyticum are in the shape of black brown sea urchins, and those of Mycoplasma homonis in the shape of fried eggs.
Main ingredients: powdered agar 3%, purified water 70%, urea 0.1%, casein peptone 2%, soy peptone 1.5%, arginine 0.5%, glucose 2%
STORAGE AND VALIDITY
1.Stored at 2~8°C.
2.The validity duration is 6 months
3.The validity duration is 1 day only once unsealing.
4.Forbidden to cryopreservation.
1.Female: Collect cervix secretion with help of vaginal speculum or amniotic fluid for culture. Urine is not recommended for culture.
2.Male: Semen, prostate secretion
3.Urethral specimens: Clean the meatus urinarius, and collect mucosal cells of the meatus with swab
4.Urine specimens: Collect the urine sediment through centrifugal, and dilute it to original volume with sterile saline.
Specimen collected above should be cultured to be slightly red in the mycoplasma liquid culture medium.
1.Use Encode mycoplasma liquid culture medium, and culture the specimen to be slightly red for spare.
2.Place mycoplasma identification agar plate in incubator for 15 minutes to make the agar surface get dry
3.Add 3 drops of the suspicious positive liquid medium, 50ul each drop respectively at 3 equal sections on the surface of the agar plate (No need to line on the surface).
4.Place the plate at room temperature for 5 minutes so as to make the specimen absorbed completely.
5.Overturn the plate, and incubate at 35~37°C in microaerophilic environment for 24~72 h.
