This kit is used for drug susceptibility test for M.tuberculosis.

 

 

Principle

 

 

  The kit adopts microplate susceptibility testing technology, which is a new MDR-TB detection methods used to determine drug susceptibility of mycobacteria with proportional method. This kit consists of liquid medium and susceptibility test board. The medium contains basic nutrients, growth stimulants and contamination inhibitors. In the medium, inorganic salts, carbon source, nitrogen source, and water work to stimulate the mycobacterium growth, whereas multi-antibiotic inhibitor suppresses the growth of foreign microbes. The test board is installed with 26 wells in which different drugs with different concentration are coated in advance in the bottom of each well. While testing, according to the growth of inoculated strains in different drug wells and control wells, the result of drug susceptibility can be reported and therefore conducive to clinical medication so as to reduce drug resistant strains.

Operating  Steps

 

  • Take out antibacterial agent, open the cap of the vial, pour out the sterile diluent  into it and shake it up.

Take out liquid medium and test board and let them warm up to room temperature before operation, then add 100ul of mixed antibacterial agent into the liquid culture medium and shake up.

Transfer 180ul of liquid culture medium into well A1 or E1 as 1/10 reference control well; And transfer 180ul of liquid culture medium into well B1 or F1 as 1/100 reference control well.

Transfer 200ul of liquid culture medium into well C1 and D1 or well G1 and H1 respectively as negative control well.

The isolated strains should be prepared to 1mg/ml by grinding turbidimetry, then inoculate 100ul of prepared suspension into the liquid culture medium and shake up; then apply 200ul of inoculated medium into the rest wells; then transfer 20ul into well A1 or E1 as 1/10 reference control well; and then transfer 20ul from well A1 or E1 and add into well B1 or F1 as 1/100 reference control well; then cover the microplate and sealing it around the perimeter. Put the test board at 35-37°C in the incubator for culture.

 

 

Result Report

 

1.     Observe the result after 7 days, if there is whitish sediment of strains on the bottom of the well, the result is positive, if not, it is negative. If negative, observe the result again after 10 days, if there is whitish sediment of strains on the bottom of the well, the result is positive, if still not, the result is negative.

 3.     Result of Drug Susceptibility: When it shows positive in positive control well (well A2 and B2 or well E2 and F2), record MIC value of each drug. MIC value is the lowest drug concentration of well without sediment in it. Put the MIC value on the report pamphlet.
